Electrometric titration of copper and zinc in biologic material.

C OLORIMETRIC ANALYSIS has been employed extensively in determination of copper and zinc in human serum and other biologic materials. Greenleaf (1) and Callan and Henderson (2) determined copper colorimetrically using dithizone and diethyldithiocarbamate, respectivelv. Zinc was determined colorinietricallv by Fischer and Leopoldi (4) and by Wolf (5), using dithizone. These procedures were evaluated by 1argerunl and Santacana (6). The methods were hampered by the time-consuming digestion, light-sensitive reagent, and extraction procedure required. Wet oxidation, described by Smith (3), is required on solid material. The method using tetraethylenepentamine (Tetren*) in potentiometric titration eliminates the above-cited difficulties and provides a rapid, accurate procedure for the determination of copper and zinc in a single sample which fulfills the clinical requirement. Martin and Reilley (7) and Johnson (8) used a dual polarized mercury electrode for the determination of trace mineral, employing tetremi as titrant. This method permits the direct determination of copper and zinc in a single sample at selected pH. The copper and zinc analysis is made on a dilute deproteinized serum sample at pH 5 and pH 9.5, respectively, titrating with Tetren potentiometrically, using dual gold polarized electrode to the respective end point.
